Overview

This drug combination is a triple therapy regimen used for the radical cure of malaria caused by *Plasmodium vivax*. It consists of an Artemisinin-based Combination Therapy (ACT)—dihydroartemisinin and piperaquine—paired with primaquine. Dihydroartemisinin provides rapid reduction of the parasite biomass in the blood, while piperaquine, a long-acting bisquinoline, provides sustained post-treatment prophylaxis and clears remaining blood-stage parasites. Primaquine, an 8-aminoquinoline, is the critical component for radical cure as it targets the dormant liver stages (hypnozoites) of *P. vivax*, thereby preventing clinical relapses. This specific regimen has been evaluated in Phase 3 clinical trials led by the University of Oxford to establish efficacy and safety in regions with chloroquine-resistant malaria.
