Overview

Dilatistimab (RO7508182, RG6234) is a bispecific T-cell engager (TCE) antibody developed by Roche for the treatment of small-cell lung cancer (SCLC) and other neuroendocrine tumors. It is engineered to simultaneously bind to Delta-like ligand 3 (DLL3), a protein frequently overexpressed on the surface of SCLC cells but with limited expression in normal tissues, and the CD3 epsilon subunit of the T-cell receptor complex. This dual binding brings T cells into close proximity with tumor cells, leading to the formation of an immunological synapse, T-cell activation, and the subsequent release of cytotoxic granules (perforin and granzymes) that induce apoptosis in the target cancer cells. Dilatistimab is currently undergoing clinical evaluation to assess its safety, tolerability, and preliminary efficacy in patients with advanced DLL3-positive malignancies.
