ES104 (dilpacimab) is a bispecific monoclonal antibody designed to simultaneously target vascular endothelial growth factor A (VEGF-A) and Delta-like ligand 4 (DLL4). This dual-targeting approach is intended to provide more robust inhibition of tumor angiogenesis compared to agents targeting VEGF alone, as DLL4-mediated Notch signaling is a key pathway involved in resistance to anti-VEGF therapies. Originally discovered by ABL Bio as ABL001, the asset was licensed to TRIGR Therapeutics (later acquired by Compass Therapeutics) and Elpiscience Biopharmaceuticals. It is currently under clinical evaluation for various solid tumors, with a particular focus on biliary tract cancer and colorectal cancer, where it has shown promising anti-tumor activity in combination with chemotherapy.
