Drug intelligence / Profile preview
Dimericine (T4N5 liposome lotion) is a topical DNA repair enzyme therapy developed by Applied Genetics Inc. (and later associated with Dermatics) for the prevention of skin cancer in high-risk individuals, particularly those with xeroderma pigmentosum (XP). The active ingredient is T4 endonuclease V, a prokaryotic DNA repair enzyme derived from the T4 bacteriophage. This enzyme is encapsulated in pH-sensitive liposomes to facilitate delivery into keratinocytes. Once inside the cell, T4 endonuclease V initiates the repair of cyclobutane pyrimidine dimers (CPDs), which are DNA lesions caused by ultraviolet (UV) radiation. By supplementing the body's natural nucleotide excision repair (NER) pathway, the drug aims to reduce the incidence of actinic keratoses and basal cell carcinomas. Although it received Orphan Drug and Fast Track designations from the FDA, development was ultimately discontinued.
