Overview

Dimethyl malonate (DiMAL or DMM) is a small molecule research compound being investigated by the University of Aarhus for its cardioprotective effects. It acts as a prodrug that is hydrolyzed to malonate, which then functions as a competitive inhibitor of succinate dehydrogenase (SDH), also known as mitochondrial complex II. By blocking succinate oxidation, the compound reduces the accumulation of succinate during ischemia and the subsequent burst of reactive oxygen species (ROS) during reperfusion, thereby mitigating ischemia-reperfusion injury. Research at Aarhus University has focused on its potential to reduce infarct size in patients undergoing heart surgery, particularly comparing effects in diabetic versus non-diabetic cardiac tissue.
