Dimethyldithiocarbamate (MDTC) is a small molecule dithiocarbamate derivative and a metabolite of the anti-alcoholism drug disulfiram. It acts as a potent inhibitor of the DNA repair protein O6-methylguanine-DNA methyltransferase (MGMT) by covalently binding to the active-site cysteine 145, leading to protein degradation. By inactivating MGMT, MDTC sensitizes glioblastoma cells to alkylating agents like temozolomide. Additionally, MDTC has been shown to inhibit NF-κB and GSTP1 signaling. Research at Texas Tech University Health Sciences Center has explored its use in glioblastoma, including nanoparticle-based delivery systems (e.g., pegylated PLGA nanoparticles) and zinc-chelated formulations.
