Overview

DiNAK-004 (also known as DF6234 or GS-6234) is a tri-specific natural killer (NK) cell engager therapy (TriNKET) developed by Dragonfly Therapeutics and licensed to Gilead Sciences. It is designed to target B7-H3 (CD276), a checkpoint molecule overexpressed in various solid tumors, including lung, prostate, and breast cancers. The TriNKET platform works by simultaneously binding to the tumor antigen (B7-H3) and activating NK cells through the engagement of both CD16a and NKG2D receptors. This dual activation of NK cells, combined with the recruitment of the adaptive immune system, aims to enhance anti-tumor activity compared to traditional monoclonal antibodies. Gilead Sciences acquired the exclusive global rights to DF6234 in 2022 to expand its oncology pipeline and is currently evaluating the asset in Phase 1 clinical trials.
