Overview

Dinitrophenyl (DNP) is a small molecule hapten-based immunotherapy agent being developed by the Chinese PLA General Hospital for the treatment of advanced, unresectable stage III or IV malignant melanoma. Functioning as a semi-antigen, DNP is administered in situ to conjugate with tumor-associated proteins, thereby creating highly immunogenic neoantigens that enhance immune recognition and overcome tumor-induced immune tolerance. This process facilitates the activation of focal T cells and induces a regional antitumor immune response, increasing cell-mediated immunity. In clinical research (NCT02372708), DNP in situ immunotherapy is utilized as an adjunct to dacarbazine chemotherapy and combined with laser therapy, a regimen that has demonstrated significant improvements in overall survival and disease-free survival for patients with skin malignant melanoma.
