DNP itself is a small molecule not immunogenic on its own, but when conjugated to proteins, it forms neo-antigens, triggering robust immune activation[4]. In clinical settings, DNP is used as a topical formulation, often combined with focal laser therapy, to induce local contact dermatitis and delayed-type hypersensitivity (DTH) in malignant melanoma patients, resulting in focal T-cell activation, increased cell-mediated immunity, and improved clinical outcomes[2][4]. The therapy aims to recruit and activate regional T cells by presenting DNP-modified tumor antigens via MHC molecules on antigen-presenting cells, eliciting a cytotoxic T cell response against the tumor.
