This combination therapy consists of the anti-GD2 monoclonal antibody dinutuximab beta (Qarziba) administered alongside the rapid COJEC induction chemotherapy regimen. Rapid COJEC is a multi-agent chemotherapy protocol comprising cisplatin, vincristine, carboplatin, etoposide, and cyclophosphamide. This specific combination is being evaluated in the SIOPEN-Pilot01 Phase Ib clinical trial, sponsored by the Princess Maxima Center for Pediatric Oncology, for the treatment of newly diagnosed high-risk neuroblastoma in pediatric patients. Dinutuximab beta works by binding to the GD2 disialoganglioside on neuroblastoma cells, inducing antibody-dependent cell-mediated cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). The addition of immunotherapy to standard induction chemotherapy aims to improve outcomes in high-risk patients by targeting residual disease and enhancing the cytotoxic effects of the chemotherapy components.
