Overview

Dinutuximab beta + GPOH chemotherapy is a combination treatment regimen currently under clinical investigation for the induction phase of newly diagnosed high-risk neuroblastoma. The regimen consists of dinutuximab beta (Qarziba), a chimeric monoclonal antibody targeting the disialoganglioside GD2, and the GPOH (German Pediatric Oncology and Hematology Group) induction chemotherapy protocol. Dinutuximab beta exerts its antitumor effect by binding to GD2 on the surface of neuroblastoma cells, thereby triggering antibody-dependent cell-mediated cytotoxicity (ADCC) and complement-dependent cytotoxicity (CDC). The GPOH chemotherapy component typically involves intensive cycles of cytotoxic agents such as cisplatin, etoposide, vindesine, cyclophosphamide, and doxorubicin. This combination is being evaluated in the Phase Ib SIOPEN-Pilot01 trial, sponsored by the Princess Maxima Center for Pediatric Oncology, to establish the safety and recommended dose of integrating anti-GD2 immunotherapy into intensive induction chemotherapy.
