Overview

**DIOS-CoVax2** is a synthetic DNA vaccine candidate developed by DIOSynVax, a University of Cambridge spin-out, using computational design to encode custom antigens targeting conserved regions of SARS-CoV-2 spike protein and related coronaviruses from genetic sequence banks, including bat-hosted strains. It elicits neutralizing antibodies and T-cell responses against critical viral docking sites while avoiding hyper-inflammatory epitopes, aiming for broad protection against current and future sarbecoviruses. The thermostable, freeze-dried plasmid DNA is delivered needle-free via intradermal jet injection (PharmaJet Tropis system) for safe, scalable use, particularly in low-resource settings; funded by UK government (£1.9M via Innovate UK), it advanced to Phase I trials in 2021 at Southampton and Cambridge sites.[1][3]
