Overview

Dioxadet is a synthetic antitumor agent from the ethylenimine group with the chemical structure 2,4-bis(1-aziridinyl)-6-(2,2-dimethyl-5-hydroxymethyl-1,3-dioxan-5-yl)amino-1,3,5-triazine. It functions as an alkylating agent, targeting DNA to interfere with cell division and causing cytotoxicity in tumor cells. Dioxadet was initially developed and clinically trialed in Russia and has been formulated into nanoforms (e.g., nanogels, silica particles, nanodiamonds) to improve selective delivery and reduce systemic toxicity. The main side effect is suppression of hematopoiesis. Its use has primarily focused on malignant neoplasms, especially brain and solid tumors, and it showed preclinical and some clinical antitumor activity. As of the most recent data, dioxadet is not currently produced or registered for use due to expiration of regulatory documentation[1][2].
