DIPG lysate is a therapeutic biological agent consisting of a processed mixture of proteins and antigens derived from diffuse intrinsic pontine glioma (DIPG) cell lines. Developed by Fundacio Sant Joan De Deu, it serves as the primary antigenic component for personalized immunotherapy, specifically autologous dendritic cell vaccines. The lysate is produced by lysing allogeneic DIPG cells (such as the HSJD-DIPG-007 line), which releases a broad spectrum of tumor-associated antigens (TAAs). When these antigens are loaded onto a patient's own dendritic cells and administered back to the patient, they prime the immune system to recognize and mount a cytotoxic T-lymphocyte response against the unique molecular profile of the brainstem tumor. This approach is designed to address the highly aggressive and infiltrative nature of DIPG, a pediatric cancer where surgical resection is not possible and conventional therapies offer limited long-term survival.
