Overview

Diphenylpyraline is a first-generation antihistamine of the diphenylpiperidine class with anticholinergic and significant sedative properties. It acts primarily as an antagonist at the histamine H1 receptor on effector cells to prevent responses mediated by histamine such as urticaria and pruritus. Diphenylpyraline also exhibits antimuscarinic effects and may provide a drying effect on nasal mucosa. Additionally, it has been found to act as a dopamine reuptake inhibitor and produces hyperactivity in rodents; it has shown utility in the treatment of Parkinsonism. The drug is marketed in Europe for allergic conditions including allergic rhinitis (hay fever) and pruritic skin disorders[1][2][5].
