Diquafosol is a small molecule pharmaceutical drug used primarily for the treatment of dry eye disease (DED) and keratoconjunctival disorders. It acts as an agonist of the P2Y2 purinergic receptor, which is expressed on conjunctival epithelial cells and goblet cell membranes. By activating these receptors, diquafosol stimulates both fluid secretion from conjunctival epithelial cells and mucin secretion from goblet cells, leading to increased tear film stability and hydration of the ocular surface. This mechanism improves symptoms in patients with dry eye by enhancing tear fluid and mucin production independently of lacrimal gland function[1][2][3][5][6]. Diquafosol is typically administered as a 3% ophthalmic solution[7].
