Overview

**Direclidine** is an investigational, orally available small molecule antipsychotic developed for the treatment of schizophrenia. It is a highly selective orthosteric agonist of the muscarinic acetylcholine M4 receptor, modulating dopaminergic neurotransmission indirectly and offering a non-dopaminergic mechanism for antipsychotic efficacy. Direclidine stems from a collaboration between Nxera Pharma and Heptares Therapeutics, was previously out-licensed to Allergan and AbbVie, and is currently being developed by Neurocrine Biosciences. Direclidine is in Phase III development for schizophrenia, having demonstrated improvement in symptoms as measured by the Positive and Negative Syndrome Scale (PANSS) in Phase II trials. It is not being actively developed for Alzheimer's disease at this time[4][5][7][10][12][13][17].
