9MW3011 is a first-in-class investigational monoclonal antibody targeting transmembrane protease serine 6 (TMPRSS6). Developed by Mabwell at its San Diego Innovation and R&D Center and licensed to Disc Medicine for development outside Greater China and Southeast Asia, it is designed to regulate iron homeostasis in vivo. The drug acts by binding specifically to TMPRSS6 on hepatocyte membranes in the liver. This interaction upregulates hepcidin expression—a key hormone that controls iron absorption and release—thereby inhibiting intestinal iron uptake and reducing serum iron levels. The primary indications under investigation are β-thalassemia and polycythemia vera; additional research includes non-transfusion dependent thalassemia, iron overload disorders, and sickle cell anemia. It has received Orphan Drug Designation (polycythemia vera) and Fast Track status from the US FDA[1][2][3][4][5][6].
