DISC-3405 (formerly MWTX-003) is a novel humanized monoclonal antibody that targets transmembrane serine protease 6 (TMPRSS6, also known as Matriptase-2). By inhibiting TMPRSS6, DISC-3405 stimulates endogenous production of hepcidin, a master regulator of iron homeostasis. Increased hepcidin reduces iron absorption and release from stores, thereby lowering serum iron levels. This mechanism is being developed to treat hematologic diseases characterized by iron overload or dysregulation, such as polycythemia vera and beta-thalassemia. The drug was in-licensed by Disc Medicine from Mabwell Therapeutics and has received both FDA Orphan Drug Designation and Fast Track status for polycythemia vera. It is currently in Phase 1 clinical trials in healthy volunteers[1][2][3][4][5][6][7].
