Overview

DISCO (Dual Interacting Subunit COmplex) CAR T cells are an experimental chimeric antigen receptor T-cell therapy designed to selectively target the α3β4 nicotinic acetylcholine receptor (nAChR). Developed by researchers at the Children's Hospital of Philadelphia, these CAR T cells utilize specialized single-chain variable fragments (scFvs) that recognize a unique epitope formed only when the α3 (CHRNA3) and β4 (CHRNB4) subunits are complexed together. This dual-subunit requirement enhances tumor selectivity, as the α3β4 complex is overexpressed in neuroblastoma, small cell lung cancer (SCLC), and retinoblastoma, but is largely absent in normal tissues. Preclinical studies have demonstrated potent cytotoxicity and improved survival in patient-derived xenograft models of neuroblastoma and retinoblastoma.
