Disitamab vedotin is an antibody-drug conjugate (ADC) consisting of a humanized IgG1 monoclonal antibody targeting the human epidermal growth factor receptor 2 (HER2), conjugated via a cleavable linker to the cytotoxic agent monomethyl auristatin E (MMAE). The drug binds specifically to HER2 on tumor cells, enabling targeted delivery and release of MMAE within these cells. MMAE disrupts microtubule polymerization, leading to cell cycle arrest and apoptosis. Disitamab vedotin was developed by RemeGen and is approved in China for HER2-overexpressing locally advanced or metastatic gastric cancer in patients who have received at least two prior systemic chemotherapy regimens. It is also under investigation for other solid tumors including urothelial carcinoma, biliary tract cancer, non-small cell lung cancer, and breast cancer[1][3][5].
