Overview

This is a combination therapy consisting of three agents: - **Disitamab vedotin** is an antibody-drug conjugate targeting human epidermal growth factor receptor 2 (HER2). It comprises a humanized monoclonal antibody (hertuzumab) linked via a cleavable valine-citrulline linker to the cytotoxic agent monomethyl auristatin E (MMAE). The drug binds HER2 on tumor cells, is internalized, and releases MMAE intracellularly to disrupt microtubules and induce cell death. Disitamab vedotin has demonstrated efficacy in HER2-positive solid tumors, including gastric and urothelial cancers[1][3][5]. - **AK104** (cadonilimab) is a tetravalent bispecific monoclonal antibody that simultaneously targets programmed cell death protein 1 (PD-1) and cytotoxic T lymphocyte-associated antigen 4 (CTLA-4), both immune checkpoint proteins. By blocking these pathways, it enhances T-cell activation against tumors while its Fc-null design reduces immune-related toxicities[7]. - **Bevacizumab** is a recombinant humanized monoclonal antibody that inhibits vascular endothelial growth factor A (VEGF-A), thereby suppressing angiogenesis required for tumor growth. The combination aims to provide synergistic antitumor effects by targeting cancer cells directly with an ADC, modulating the immune response through dual checkpoint inhibition, and inhibiting tumor angiogenesis.
