This combination therapy consists of disitamab vedotin, cisplatin, and toripalimab. Disitamab vedotin is a HER2-targeted antibody-drug conjugate (ADC) composed of a humanized anti-HER2 monoclonal antibody conjugated to the microtubule-disrupting agent monomethyl auristatin E (MMAE) via a protease-cleavable linker. Cisplatin is a platinum-based chemotherapeutic agent that induces DNA cross-linking, leading to apoptosis. Toripalimab is a humanized IgG4 monoclonal antibody that targets the programmed cell death protein 1 (PD-1) receptor, blocking its interaction with PD-L1 and PD-L2 to restore T-cell mediated anti-tumor immune responses. This triplet regimen is being investigated, particularly in China by institutions like Beijing Anzhen Hospital, as a neoadjuvant or systemic treatment for patients with HER2-expressing urothelial carcinoma to maximize tumor shrinkage and improve survival outcomes through synergistic targeted cytotoxicity and immune activation.
