Disitamab vedotin (RC48-ADC) is a HER2-targeted antibody-drug conjugate composed of a humanized anti-HER2 monoclonal antibody (hertuzumab) linked to the cytotoxic agent monomethyl auristatin E (MMAE) via a cleavable linker. It selectively binds to HER2-expressing tumor cells, is internalized by endocytosis, and releases MMAE intracellularly to disrupt microtubule formation and induce apoptosis. The drug has demonstrated efficacy in HER2-positive and HER2-low cancers such as metastatic urothelial carcinoma, gastric cancer, and breast cancer[2][4][5]. Gemcitabine is a nucleoside analog used as chemotherapy that inhibits DNA synthesis. The combination of RC48-ADC with gemcitabine aims to enhance antitumor activity through complementary mechanisms—targeted delivery of cytotoxin by RC48-ADC and broad cytotoxicity from gemcitabine[1]. Disitamab vedotin was developed by RemeGen for the treatment of various solid tumors.
