disitamab vedotin + penpulimab + cisplatin is a combination therapy regimen being investigated for the treatment of HER2-positive advanced gastric cancer. The regimen consists of disitamab vedotin (RC48), a HER2-targeted antibody-drug conjugate (ADC) developed by RemeGen that delivers the microtubule-disrupting agent monomethyl auristatin E (MMAE); penpulimab (AK105), a humanized PD-1 monoclonal antibody developed by Akeso; and cisplatin, a platinum-based chemotherapeutic agent that induces DNA cross-linking. This triple combination aims to leverage the targeted cytotoxicity of the ADC, the immune-potentiating effects of the checkpoint inhibitor, and the broad antineoplastic activity of cisplatin to improve outcomes in patients with HER2-overexpressing gastric or gastroesophageal junction cancers. It is currently being evaluated in Phase 2 clinical trials sponsored by Henan Cancer Hospital.
