Overview

**Disitamab vedotin + RC148** is an investigational combination therapy comprising two novel biologic drugs:\n- **Disitamab vedotin** (also known as RC48) is an antibody-drug conjugate (ADC) directed against the human epidermal growth factor receptor 2 (HER2), composed of a humanized anti-HER2 antibody (hertuzumab) conjugated to the microtubule inhibitor monomethyl auristatin E (MMAE) via a cleavable valine-citrulline linker. Disitamab vedotin selectively binds HER2 on tumor cells, is internalized, and releases MMAE intracellularly to disrupt microtubules, resulting in apoptosis of HER2-expressing cancer cells[4]. \n- **RC148** is a bispecific antibody targeting both programmed cell death protein 1 (PD-1) and vascular endothelial growth factor (VEGF), functioning as an immunomodulator with dual immune checkpoint and angiogenesis inhibition properties; it was developed using RemeGen's proprietary bispecific antibody technology[1][2][3][5][7].\n\nThis combination is under clinical investigation for locally advanced or metastatic solid tumors, in particular for HR-negative, HER2-low expressing unresectable locally advanced or metastatic breast cancer. Multiple clinical trials are running or recruiting as of 2025[3][6][8]. Both agents are administered by intravenous infusion.
