Overview

Disitertide is a synthetic peptide and transforming growth factor-beta 1 (TGF-β1) inhibitor designed to block the interaction of TGF-β1 with its receptor. It acts as a TGF-β1 antagonist peptide and has demonstrated antifibrogenic activity in preclinical models by reducing binding of TGF-β1 to its receptors and inhibiting downstream signaling pathways such as SMAD2 phosphorylation. Disitertide also induces apoptosis and inhibits PI3K. The drug has been investigated primarily for fibrotic diseases, including systemic scleroderma and skin fibrosis, with orphan drug designation for systemic scleroderma. It has shown potential in reducing fibrosis in animal models and improving scar morphology in vivo. Development was led by Digna Biotech and the University of Navarra, with involvement from ISDIN and Avadel Pharmaceuticals[1][2][3][4][5][7].
