Overview

Distearoylphosphatidylethanolamine-polyethylene glycol-rabies virus glycoprotein peptide (DSPE-PEG-RVG) is a functionalized chimeric lipid conjugate designed for targeted drug delivery to the central nervous system. It consists of a phospholipid tail (DSPE) for membrane anchoring, a polyethylene glycol (PEG) spacer to enhance stability and circulation time, and a 29-amino acid peptide derived from the rabies virus glycoprotein (RVG29). The RVG peptide specifically binds to the nicotinic acetylcholine receptor (nAChR) expressed on the blood-brain barrier (BBB) endothelial cells and neurons, facilitating receptor-mediated transcytosis. This allows the delivery of various therapeutic cargoes, such as siRNA, chemotherapeutics, or imaging agents, across the BBB into the brain parenchyma. It is widely utilized in the development of brain-targeted liposomes, micelles, and other lipid-based nanoparticles for treating neurological disorders and glioblastoma.
