DJ-1 lentivirus is a gene therapy construct consisting of a lentiviral vector designed to overexpress the DJ-1 protein (encoded by the PARK7 gene). DJ-1 is a multifunctional protein with antioxidant and chaperone activities, often associated with neuroprotection. In preclinical models of Alzheimer's disease (such as 5XFAD mice), hippocampal injection of this lentivirus has been shown to reduce amyloid-beta (Aβ) deposition, decrease oxidative stress (by activating the Nrf2 pathway), and inhibit neuronal pyroptosis (by suppressing the NLRP3 inflammasome and caspase-1 activation). This leads to improved cognitive function and reduced brain tissue damage.
