DJ-1 siRNA is a small interfering RNA specifically designed to silence expression of the DJ-1 gene (also known as PARK7), a multifunctional protein implicated in cancer cell survival, resistance to chemotherapy, oxidative stress defense, and suppression of apoptosis. By targeting DJ-1 mRNA, DJ-1 siRNA downregulates DJ-1 protein expression, impairing cancer cell proliferation, migration, and survival. Mechanistically, DJ-1 siRNA exerts anti-cancer effects through inhibition of the Akt signaling pathway, restoration of p53-mediated apoptosis and cell cycle arrest, reduction of Nrf2-mediated antioxidant response, and increased sensitivity to chemotherapeutic agents such as cisplatin or adriamycin. These properties have been evaluated in preclinical cancer models, particularly for platinum-resistant ovarian cancer and multidrug-resistant breast cancer, using various targeted or nanoparticle-based delivery systems[1][2][3].
