DJ101 is a novel small molecule tubulin inhibitor that specifically targets the colchicine binding site on the beta-tubulin subunit. Developed by researchers at the University of Tennessee Health Science Center, it acts as a microtubule destabilizing agent, disrupting microtubule dynamics and polymerization. DJ101 has demonstrated potent in vitro activity against a panel of metastatic melanoma cell lines, including those with clinically relevant mutations, and effectively circumvents drug efflux pumps that often limit the efficacy of other antimitotic agents like taxanes. In preclinical studies, DJ101 showed significant tumor growth inhibition in human melanoma xenograft models and reduced metastatic potential in lung metastasis models, while maintaining a favorable safety profile with minimal off-target effects.
