Overview

DK-233 is an experimental fluoroquinolone antibiotic developed by Daiichi Sankyo (formerly Daiichi Pharmaceutical) for the treatment of tuberculosis. As an 8-methoxyquinolone, it was designed to exhibit enhanced potency against mycobacteria and Gram-positive pathogens while minimizing the selection of resistant mutants. The drug's mechanism of action involves the inhibition of DNA gyrase and topoisomerase IV, which are critical for maintaining DNA topology during replication and transcription. Preclinical evaluations demonstrated that DK-233 has significant in vitro activity against both drug-susceptible and multi-drug resistant (MDR) strains of *Mycobacterium tuberculosis*, as well as efficacy in murine models of infection. Despite its promising antibacterial profile, the development of DK-233 appears to have been discontinued in favor of other candidates or due to strategic shifts in the developer's pipeline.
