DK-CLIC-1901 is an autologous, second-generation chimeric antigen receptor (CAR) T-cell therapy targeting the CD19 antigen. Developed by Rigshospitalet in Denmark, it is designed for the treatment of relapsed or refractory B-cell malignancies, including acute lymphoblastic leukemia (ALL) and various subtypes of non-Hodgkin lymphoma (NHL). The CAR construct incorporates an anti-CD19 single-chain variable fragment (scFv) linked to a 4-1BB (CD137) costimulatory domain and a CD3-zeta signaling domain. The therapy is produced using an automated manufacturing platform at the hospital's own facility, aiming to provide a locally produced, point-of-care alternative to commercial CAR-T products. It is currently being evaluated in Phase 1/2 clinical trials for patients with CD19-positive B-cell cancers who have failed standard therapies.
