**DK210 (EGFR)** is a tumor-targeted dual immunocytokine developed by Deka Biosciences using the Diakine platform, coupling wild-type interleukin-2 (IL-2) to a high-affinity variant of Epstein-Barr viral interleukin-10 (IL-10) via a single-chain variable fragment (scFv) scaffold that binds epidermal growth factor receptor (EGFR). This design enhances IL-2 potency while mitigating toxicity through IL-10's balancing effect, promoting effector T cell and NK cell expansion, IFNγ production, and eosinophilia without cytokine release syndrome (CRS), vascular leak, or regulatory T cell accumulation. It is self-administered subcutaneously three times weekly and is being evaluated in a phase 1 trial (NCT05704985) for advanced EGFR-expressing solid tumors, showing stable disease in patients with colorectal, pancreatic, and non-small cell lung cancers, with further monotherapy and combination studies planned in renal cell carcinoma and non-small cell lung cancer.[1][2][5][10]
