Overview

DK210 + chemotherapy refers to a combination regimen of DK210, a next-generation tumor-targeted dual immunocytokine, with standard chemotherapy agents. DK210 (also referred to as DK210 (EGFR)) consists of wild-type interleukin-2 (IL-2) coupled to an Epstein Barr Virus (EBV) IL-10 variant, both fused onto a single-chain fragment variable (scFv) that targets the epidermal growth factor receptor (EGFR). The molecule is designed to deliver immune-activating cytokines specifically to EGFR-overexpressing tumor cells, enhancing immune activation (notably effector T cell and NK cell expansion) while reducing regulatory T cell (Treg) expansion and limiting cytokine release syndrome (CRS)-related toxicity. The chemotherapy component refers to the addition of standard-of-care cytotoxic drugs, although the precise agents are not universally specified and may vary by tumor type or protocol. This combination is being explored for various advanced solid tumors characterized by EGFR overexpression, with the aim of enhancing anti-tumor efficacy and improving safety over existing immunotherapy and chemotherapy regimens[1][2][3][5][6].
