Overview

DKF-457 is an orally administered small molecule generic candidate of lobeglitazone (originally developed and marketed as Duvie by Chong Kun Dang Pharmaceutical), currently under development by Dongkook Pharmaceutical for the treatment of type 2 diabetes mellitus. As a thiazolidinedione (TZD) class antidiabetic agent, its mechanism of action involves the selective activation of peroxisome proliferator-activated receptor gamma (PPAR-γ), a nuclear receptor that regulates the transcription of genes involved in glucose and lipid metabolism. Activation of PPAR-γ enhances insulin sensitivity in key target tissues, including adipose tissue, skeletal muscle, and the liver, thereby promoting glucose uptake and reducing hepatic glucose output. In South Korea, Dongkook Pharmaceutical has completed bioequivalence studies for DKF-457, positioning the drug for regulatory approval and subsequent market launch.
