Drug intelligence / Profile preview
DKK1-A2 CAR-T cells are chimeric antigen receptor (CAR) T cells engineered to target the Dickkopf-1 (DKK1) peptide in the context of the human HLA-A2 complex (DKK1-A2 complex). Developed by researchers at the Houston Methodist Research Institute and Houston Methodist Neal Cancer Center, these CAR-T cells utilize a T-cell receptor-like monoclonal antibody (such as the C2 clone) to recognize the tumor-associated antigen DKK1-A2 complex, which is highly expressed on various hematologic malignancies (including multiple myeloma, acute myeloid leukemia, and chronic myeloid leukemia) and solid tumors (such as pancreatic, lung, and breast cancers) but absent in most normal tissues. Preclinical studies have demonstrated that DKK1-A2 CAR-T cells, particularly when polarized under Th9 conditions to enhance persistence and cytolytic activity, specifically lyse HLA-A2+ DKK1+ tumor cells in vitro and effectively eradicate established tumors in vivo without significant on- or off-target toxicities.
