DKK1-A2 mAbs are a series of therapeutic monoclonal antibodies that target the Dickkopf-1 (DKK1) peptide (specifically the P20 peptide) presented in the context of the HLA-A*0201 (HLA-A2) molecule on the surface of malignant cells. Unlike conventional DKK1-targeting antibodies that bind to the secreted form of the protein, these antibodies are designed to recognize the peptide-MHC complex, enabling the specific targeting of DKK1-expressing tumor cells while sparing normal tissues. The antibodies demonstrate anti-tumor activity through the direct induction of apoptosis via the caspase-9 cascade, as well as by mediating immune-effector functions such as complement-dependent cytotoxicity (CDC) and antibody-dependent cellular cytotoxicity (ADCC). Preclinical studies conducted by researchers at The Houston Methodist Research Institute have shown efficacy in various hematologic and solid tumors, including multiple myeloma, lymphoma, leukemia, and breast, pancreatic, and prostate cancers.
