DKK1-DNA vaccine is an investigational DNA vaccine designed to induce an immune response against Dickkopf-1 (DKK1), a secreted antagonist of the Wnt signaling pathway that is overexpressed in multiple myeloma and other cancers. The vaccine functions by encoding the DKK1 antigen in a DNA plasmid, which is then delivered to host cells to elicit humoral and cellular anti-tumor immune responses, particularly cytotoxic T lymphocyte (CTL) and T-helper responses. Preclinical studies in murine models have demonstrated that the DKK1-DNA vaccine can both protect against myeloma development and provide therapeutic benefit in established disease, largely mediated by DKK1-specific CD4+ and CD8+ T cell responses, with further enhancement by CpG adjuvant and immunomodulatory antibodies (such as B7H1 blockade or OX40 agonism). Fusion constructs, such as human DKK1 fused with human HSP70 (hDKK1-hHSP70 DNA vaccine), have also been developed to enhance immunogenicity and anti-tumor efficacy. The approach represents a novel cancer immunotherapy strategy targeting a shared tumor-associated antigen[2][3][5][8].
