Overview
The DKK1-loaded autologous dendritic cell vaccine is an investigational cell-based immunotherapy developed by the Case Comprehensive Cancer Center for the treatment of plasma cell disorders, including monoclonal gammopathy of undetermined significance (MGUS), smoldering multiple myeloma, and stable multiple myeloma. The vaccine is produced by harvesting a patient's own dendritic cells and exposing them to Dickkopf-1 (DKK1) peptides. DKK1 is a secreted protein and Wnt signaling pathway inhibitor that is highly expressed in multiple myeloma cells but has minimal expression in most normal adult tissues, making it a suitable tumor-associated antigen. Once re-infused, these primed dendritic cells present DKK1 antigens to the patient's T cells, aiming to stimulate a targeted immune attack against myeloma cells. The therapy is currently being evaluated in pilot clinical trials to assess safety and preliminary efficacy.
