Overview
A DNA-based vaccine designed to induce immunity against Dickkopf-1 (DKK1), a secreted inhibitor of the canonical Wnt signaling pathway that is upregulated in several bone-related pathologies (especially rheumatoid arthritis and multiple myeloma)[1][2][3][8]. The vaccine utilizes multiple B cell epitope fragments from DKK1, linked together in a synthetic gene construct, and is typically delivered via plasmid DNA for intramuscular immunization. This approach neutralizes DKK1 activity, leading to reduced bone erosion and anti-tumor effects in mouse models. The vaccine stimulates both humoral and cellular immune responses, including the production of neutralizing antibodies against DKK1 and DKK1-specific CD4+ and CD8+ T cells[1][2][3].
