Overview

DKK678 is a novel recombinant scorpion peptide designed through bioinformatic modeling and molecular docking to selectively target the voltage-gated sodium channel subtype Nav1.5. It was developed to address skeletal muscle inflammation and immune dysfunction associated with sepsis. By inhibiting Nav1.5, DKK678 suppresses the activation of the NF-κB signaling pathway, thereby reducing the production of pro-inflammatory cytokines such as IL-6 and TNF-α, as well as the expression of adhesion molecules like VCAM-1 and gap junction proteins like Cx43. In preclinical models of sepsis (cecal ligation and puncture), DKK678 demonstrated the ability to ameliorate muscle damage and preserve the structural integrity of immune organs like the thymus and spleen without affecting the Nav1.4 subtype, which is critical for normal muscle contraction.
