Overview
DKN-01 + gemcitabine + cisplatin is an investigational combination therapy comprising DKN-01, a humanized IgG4 monoclonal antibody targeting Dickkopf-related protein 1 (DKK1), with the chemotherapeutic agents gemcitabine and cisplatin. DKN-01 neutralizes DKK1, a secreted modulator of Wnt signaling that is implicated in tumor growth, metastasis, angiogenesis, and immunosuppression. By binding and removing DKK1, DKN-01 aims to reduce cell proliferation, block CKAP4/PI3K/AKT signaling, suppress myeloid-derived suppressor cells (MDSCs), reduce angiogenesis, and enhance natural killer (NK) cell activation. Gemcitabine is a nucleoside analog that inhibits DNA synthesis, and cisplatin is a platinum-based DNA crosslinker that induces apoptosis. The combination has been studied in unresectable or metastatic biliary tract cancer (BTC) and other advanced tumors.
