DKO CD19CAR T-cells are an experimental, "off-the-shelf" (allogeneic) chimeric antigen receptor (CAR) T-cell therapy. These cells are engineered from healthy third-party donors and undergo CRISPR/Cas9-mediated double knockout (DKO) of the T-cell receptor (TCR) and CD45 (PTPRC). The TCR knockout is intended to prevent graft-versus-host disease (GVHD), while the CD45 knockout is a platform-specific modification used in studies comparing them to CD45-targeted CAR-T cells. In research settings, such as those conducted by University College London and the University of Birmingham, DKO CD19CAR T-cells have been utilized as a negative control in xenogeneic models of acute myeloid leukemia (AML) to demonstrate the specificity and efficacy of other DKO CAR-T therapies, as AML blasts typically do not express the CD19 antigen.
