Overview

DKY709 (also known as NVP-DKY709) is an orally active, selective molecular glue degrader targeting the transcription factor IKZF2 (also called Helios), developed for cancer immunotherapy. It functions by modulating the E3 ubiquitin ligase complex to promote ubiquitination and subsequent proteasomal degradation of IKZF2. By degrading Helios in regulatory T cells (Tregs), DKY709 suppresses their function and enhances anti-tumor immune responses. This mechanism is designed to improve the efficacy of cancer immunotherapies by reducing tumor-induced immune suppression. The drug was developed by Novartis Institutes for Biomedical Research and has been evaluated in phase 1 clinical trials as a single agent and in combination with other immunotherapies for advanced solid tumors, including non-small cell lung cancer (NSCLC), melanoma, nasopharyngeal carcinoma (NPC), colorectal cancer, and other advanced malignant solid neoplasms[1][3][4][5][6].
