Overview

DL-PDMP (DL-threo-1-Phenyl-2-decanoylamino-3-morpholino-1-propanol) is a small molecule inhibitor of ceramide glucosyltransferase (UGCG), also known as glucosylceramide synthase (GCS). This enzyme catalyzes the initial step in the biosynthesis of glucosylceramide-based glycosphingolipids (GSLs) by transferring glucose from UDP-glucose to ceramide. By inhibiting this pathway, DL-PDMP leads to a reduction in complex glycosphingolipids and a concomitant accumulation of intracellular ceramide, which acts as a potent pro-apoptotic signaling molecule. In oncology research, DL-PDMP has been identified as a potential therapeutic agent for prostate cancer through drug repositioning strategies, where it has demonstrated the ability to reverse malignant gene expression profiles, induce apoptosis, and suppress colony formation in cancer cells while exhibiting low toxicity to normal epithelial cells.
