Drug intelligence / Profile preview
DL-threo-1-phenyl-2-palmitoylamino-3-morpholino-1-propanol (DL-threo-PPMP) is a synthetic small-molecule ceramide analog and a racemic mixture of D-threo and L-threo stereoisomers. It acts as a potent inhibitor of glucosylceramide synthase (ceramide glucosyltransferase), the enzyme responsible for catalyzing the transfer of glucose to ceramide, which is the first committed step in glycosphingolipid biosynthesis. By inhibiting this pathway, DL-threo-PPMP is widely used as a research tool to investigate the roles of glycosphingolipids in cell growth, apoptosis, autophagy, and differentiation. Additionally, DL-threo-PPMP has been shown to inhibit sphingomyelin synthase activity in *Plasmodium falciparum*-infected erythrocytes, blocking parasite growth and suggesting potential antimalarial applications. In preclinical models, DL-threo-PPMP has also demonstrated the ability to promote the expansion of erythroid-biased progenitors and mitigate radiation-induced anemia by fostering macrophage-hematopoietic stem cell interactions.
