Overview

DLE102 (also referred to as dlE102) is a murine oncolytic adenovirus developed as a preclinical surrogate for human oncolytic virotherapies, specifically those within the Celyvir platform. It is a modified version of the murine adenovirus type 1 (MAdV-1), engineered to selectively replicate in and lyse cancer cells. In research settings, DLE102 is frequently delivered using mesenchymal stem cells (MSCs) as vehicles (OAd-MSCs) to enhance tumor homing and protect the virus from immune neutralization. This therapeutic approach has demonstrated significant antitumor efficacy in immunocompetent mouse models of melanoma and renal adenocarcinoma. The mechanism of action involves direct oncolysis and the induction of a pro-inflammatory tumor microenvironment, characterized by increased infiltration of natural killer (NK) cells, tumor-associated macrophages (TAMs), and tumor-infiltrating lymphocytes (TILs), alongside a reduction in T-cell exhaustion markers such as PD-1.
