dLGG is a **phyto-glyceroglycolipid**, specifically 1,2-di-O-α-linolenoyl-3-O-β-galactopyranosyl-sn-glycerol, isolated from the medicinal plant Crassocephalum rabens (Asteraceae)[2]. Preclinical studies show dLGG inhibits **melanoma brain metastasis** in mice by reprogramming the tumor microenvironment, notably impeding assembly of M2-like macrophages and melanoma cell infiltration and growth in brain tissue[2]. dLGG induces anti-proliferative, anti-angiogenic, and pro-apoptotic effects against melanoma brain-seeking cells, downregulates PD-L1, Bcl-2, VEGF, MEK, ERK, c-Jun, Src, and STAT3[2]. In addition, dLGG promotes secretion of oxylipins (notably 9,10-/12,13-epoxyoctadecenoic acids) that help drive macrophage polarization toward the anti-tumor M1-phenotype. It shows synergy with liposomal doxorubicin (Lip-DOX) and bevacizumab (Avastin), further suppressing brain metastatic melanoma in murine models[2].
