DLL3-CAR-NK cells are genetically engineered natural killer (NK) cells that express a chimeric antigen receptor (CAR) targeting delta-like ligand 3 (DLL3), a protein highly expressed on the surface of small cell lung cancer (SCLC) and other neuroendocrine tumors but minimally present in normal tissues. The CAR construct enables NK cells to specifically recognize and kill tumor cells expressing DLL3. Preclinical studies have demonstrated that these modified NK-92 cell lines exhibit potent cytotoxicity against SCLC both in vitro and in vivo, leading to significant tumor regression with favorable safety profiles. A phase I clinical trial is currently underway to evaluate the safety and efficacy of this therapy in patients with relapsed or refractory extensive-stage SCLC[1][2][5].
